New approaches to radio- and chemosensitization using proton pump inhibitors through the lens of tumor cell microenvironment patterns

Background. Chaotic and improper angiogenesis in the tumor tissue causes deficit O2 perfusion tumors, a decreased capacity to deliver nutrients remove metabolism products from cells, creating preconditions for formation of hypoxic alkaline microenvironment which drives progression, invasion metastases. Recent studies have shown that use proton pump inhibitors (PPIs) increases sensitivity cells are resistant cytotoxic drugs radiation therapy.
 Purpose – provide insight into modern data on possibilities overcoming radio- chemoresistance.
 Materials methods. The selection publications was performed such databases as Pubmed, eBook Business Collection, Clinical Key Elsevier, Cochrane Library ways chemoresistance using PPIs were described. At first stage, search literature sources carried out by following keywords: radioresistance, inhibitors, cell microenvironment. second abstracts articles studied, those did not match criteria our work excluded. third full texts selected studied relevance eligibility be included references.
 Results discussion. modulation acidic is prospective direction antitumor treatment. abnormal pH-gradient between extracellular environment cytoplasm mediated different ion/proton pumps, including vacuolar H+-ATPase, Na+/H– exchangers, monocarboxylate transporters, carbonic anhydrases, etc. Several revealed inhibit only H+/K+-ATPase gastric parietal but also H+-ATPase (V-ATPase) overexpressed cells. addition treatment regimen has demonstrated effectiveness processes localization, colorectal cancer, ovarian lung pancreatic prostate breast cancer.
 Conclusions. obtained analysis indicate potential conducting preclinical clinical trials new chemosensitizing Ukraine.
